DDT (ELISA Kit)

Intended Use

The EnviroLogix DDT Plate Kit Is A Quantitative Laboratory Assay For The Detection Of Total DDT Pesticide Residues (Sum Of P,P’-DDT, -DDE, And -DDD) In Soil, With An Assay Range From 0.025 To 0.875 Parts Per Million (Ppm).

General

 

The EnviroLogix DDT Plate Kit is a competitive Enzyme-Linked ImmunoSorbent Assay (ELISA).  In the test, DDT pesticide residues in the sample compete with enzyme (horseradish peroxidase) labeled DDT for a limited number of antibody binding sites on the inside surface of the test wells.  After a simple wash step, the outcome of the competition is visualized with a color development step.  As with all competitive immunoassays, sample concentration is inversely proportional to color development.

Darker color = Lower concentration

Lighter color = Higher concentration

Materials Provided: Materials Required But Not Provided:
  • 12 x
    		•  Strips of 8 Antibody-Coated Wells each, in plate frame 1 vial of Negative Control (methanol).
  •   1 x
    		•  Vial of “0.025 ppm” DDT Calibrator (actual concentration 5 ppb in methanol*).
  •   1 x
    		• Vial of “0.1 ppm” DDT Calibrator (actual concentration 20 ppb in methanol*).
  •   1 x
    		•  Vial of “0.875 ppm” DDT Calibrator (actual concentration 175 ppb in methanol*).
  •   1 x
    		• Bottle of DDT Assay Diluent.
  •   1 x
    		•  Bottle of DDT-enzyme Conjugate Concentrate (100x).
  •   1 x
    		•  Bottle of Substrate.
  •   1 x
    		•  Bottle of Stop Solution.
    * The reason for the difference between the stated and actual calibrator concentrations is to account for the 1:5 dilution incurred during sample extraction.
  		• Balance For Weighing Samples
  		• EnviroLogix Soil Extraction Kit, or suitable sealable vessels for sample extraction.
  		• Methanol for sample extraction
  		• Positive displacement pipetter with disposable tips and plungers for dispensing of calibrators and sample extracts (capable of delivering 20 microliters (µL)).
  		• Disposable Tip, Adjustable Air Displacement Pipette(s) which will measure 120, 60 and 100 µL.
  		• Marking Pen (indelible).
  		• Tape or Parafilm.
  		• Timer (1 hour and 30 minutes).
  		• Cool Tap or Distilled Water for rinsing wells.
  		• Microtiter Plate Reader or strip reader.
  		• Microtiter Plate Washer (optional).
  		• Multi-Channel Pipette that will measure 120, 60 and 100 µL (optional).
  		• Orbital Plate Shaker (optional).
